Es ist ein hochmaligner Tumor der Blutgefäße, der von deren Endothelzellen abstammt. Aufgrund des aggressiven Verhaltens des Tumors ist eine kurative Therapie in der Regel nicht möglich, generell ist die Prognose schlecht. Nach Diagnose der Erkrankung leben nur wenige der Patienten länger als ein Jahr. Methoden zur Früherkennung gibt es derzeit nicht. Ziel dieser Arbeit war es ein in vitro Modell des malignen Hämangioendothelioms zu etablieren. Hierfür sollten Zellen des Tumors isoliert, identifiziert und charakterisiert werden. Das Modell soll einerseits dazu dienen neue Erkenntnisse zur zellulären Entstehung dieses hochmalignen Tumors zu gewinnen und andererseits künftig die Prüfung neuer Chemotherapeutika mit einem Minimum an Tierversuchen zu ermöglichen. Da die Milz bevorzugte und häufigste Entstehungslokalisation des Tumors ist, wurde neoplastisch entartetes Milzgewebe mehrerer Hunde in vitro kultiviert. Zum Vergleich wurden auch Endothelzellen aus einer gesunden Milz isoliert. Es wurde eine Strategie zur Identifizierung der Zellen erarbeitet. Diese setzte sich aus folgenden Arbeitsschritten zusammen: 1. dem immunzytochemischen Nachweis der endothelzellspezifischen Marker vWF, CD31, Tie-2 und CD51/61; 2. dem Nachweis bestimmter Lektine; 3. der Analyse des Wachstumsmusters der Zellen. Isoliert und in vitro kultiviert wurden Endothelzellen aus neoplastisch entartetem Gewebe der Milz von drei Hunden und der unveränderter Milz eines vierten Hundes. Die Zellen aus den tumorös entarteten caninen Milzen konnten bis zu 145 Tagen reproduzierbar kultiviert werden. Die Zellen, die aus einer unveränderten caninen Milz isoliert waren, konnten nur 80 Tage lang in vitro kultiviert werden. It is a highly malignant neoplasm of vascular endothelial origin. Because of the aggressive behaviour of the tumour curative therapy is not possible and in general prognosis is poor. After diagnosis only a few of the patients live longer than one year. There are currently no methods of early diagnosis. The purpose of this study was to establish an in vitro model of the malignant hemangioendothelioma. Cells of the tumour should be isolated, identified and characterised. The resulting model should increase knowledge on the cellular origin of this highly malignant tumour and allow the testing of chemotherapeutics with a minimum of animal experiments in the future. Because the spleen is a preferred and the most frequent origin of tumour localisation, transformed splenic tissue of several dogs was cultivated in vitro. Endothelial cells from a healthy spleen were isolated as well. A strategy was set up to identify the cells. This consisted of the following steps: 1\\. immunocytochemical labelling with specific markers for endothelial cells: vWF, CD31, Tie-2 and CD51/61; 2. the labelling with certain lectins; 3. the analysis of the growth pattern of the cells. Endothelial cells were isolated and cultivated in vitro from transformed splenic tissue of three dogs and the healthy spleen of one dog. Cells taken from neoplastic canine spleens could be cultivated reproducibly up to 145 days. Cells from the healthy canine spleen could be cultivated 80 days in vitro only. During in vitro cultivation differences were observed between the cultures from the malignant hemangioendothelioma and the culture from the healthy spleen.
